Prifddinas a dinas fwyaf Lesotho yw Maseru. Fe'i lleolir yng ngorllewin y deyrnas, ger y ffin â De Affrica.

Yn gorwedd ar lan Afon Mohokare, Maseru yw'r unig ddinas sylweddol yn Lesotho, gyda phoblogaeth o tua 227,880 (2006). Sefydlwyd y ddinas fel gwersyll heddlu a chafodd ei chyhoeddi yn brifddinas Lesotho pan ddaeth y wlad honno yn brotectoriaeth dan ofal Prydain yn 1869. Pan enillodd Lesotho ei hannibyniaeth yn 1966, parhaodd Maseru yn brifddinas y wladwriaeth newydd. Ystyr yr enw yn yr iaith leol yw "(Lle) Tywodfaen Coch".
